摘要
目的 :探讨氟砷联合作用对大鼠子代各器官组织超微结构的影响。 方法 :采用两代一窝繁殖试验的方法 ,对亲代接触不同剂量氟砷后 ,其子代各器官组织超微结构进行了电镜观察。并对其子代肝细胞和心肌细胞内的线粒体进行了体视学定量研究。 结果:各染毒组大鼠子代各器官组织超微结构均有一定的改变 ,可见细胞核形不规则 ,异染色质增多 ,核仁边集 ;线粒体肿胀及空泡变性 ,粗面内质网脱颗粒 ,滑面内质网扩张。结果中、高剂量组Vvm、Svm、Qm、Num明显高于对照组 (P<0 .0 5 ) ,说明染毒组肝、心线粒体均有不同程度的增生。结论:氟砷可以通过胎盘屏障或是哺乳方式对大鼠子代各器官组织超微结构有一定的影响。
Objective: To explore the joint effects of fluoride and arsenic on the organs and tissues of the rat offsprings. Methods: With two-generation nest reproductive test, the heart、 liver、 kidney、 brain、testis and ovary of the rat offsprings, whose parents exposed to different doses of fluoride and arsenic, were observed under the electron microscope and the mitochondrion of the cardiac muscle and hepotocytes were determined by morphometry. Results: There were some pathological changes in the offspring organs and tissues among all exposed groups, such as the irregularities of nuclei shape、 heterochromatin/euchromatin ratio increasing、 nucleolar margination、 mitochondrion hyperplasia、 polyribosome droping from rough endoplasmic reticulum、 smooth endoplasmic reticulum expending et al; and the Vvm、 Svn、 Qm and Num of mitochondrion in cardiac muscle and hepatocyes in middle and high groups were differently higher than control groups (P<0.05); it appeared mitochondrion hyperplasia in cardiac muscle and hepatocyes. Conclusion: It suggests that fluoride and arsenic affect the offspring's organs and tissues on ulstrastrusture of rats through breast-feeding and penetrating the barries of placenta.
