摘要
目的:探讨口腔粘膜癌前病变及口腔鳞状细胞癌发生与发展过程中 ,p5 3蛋白、p16蛋白和 PCNA的表达及意义。方法:应用免疫组织化学方法对 2 0例口腔扁平苔藓、2 0例口腔白斑、2 0例口腔鳞状细胞癌分别进行检测。结果:(1)口腔鳞状细胞癌组织中突变型 p5 3蛋白阳性率为 90 .0 %,明显高于口腔白斑 (10 .0 %)和口腔扁平苔藓 (0 .0 %) (P <0 .0 0 5 )。(2 )口腔鳞状上皮细胞癌组织中 p16蛋白阳性率为 35 .0 %,明显低于口腔白斑 (70 .0 %)和口腔扁平苔藓 (90 .0 %) (P <0 .0 0 5 )。 (3) PCNA的阳性率在口腔鳞状细胞癌组织中为 90 .0 %,明显高于口腔白斑(10 .0 %)和口腔扁平苔藓 (0 .0 %) (P <0 .0 0 5 )。 结论:p5 3基因突变使 p5 3蛋白过度表达、p16蛋白表达缺失和PCNA的过度表达在口腔鳞状上皮细胞癌的发生与发展过程中起着十分重要的作用。
Objective: To investigate the relationship of expression between p53、p16 protein and PCNA and their significance during the development and progression of oral precancerous lesion, and squamous cell carcinoma. Methods: Expression of p53、p16 protein and PCNA were detected in oral lichen planus (OLP) 20 cases, oral leukoplakia (OL) 20 cases and oral squamous cell carcinoma (OSCC) 20 cases by immunohistochemical techniques. Results: (1) The positive rate of p53 protein was higher in OSCC (90.0%) than in OL (10.0%) and OLP (0.0%) (P<0.005), (2) The positve rate of p16 protein was lower in OSCC (35.0%) than in OL (70.0%) and OLP (90.0%) (P<0.005). (3) The positive rate of PCNA was higher in OSCC (90.0%) than in OL(10.0%) and OLP (0.0%) (P<0.005) . Conclusion: p53 gene mutation resulting in over-expresion of p53 protein, the low-expression of p16 protein and over-expression of PCMA may play important roles in oral squamous cell carcinoma development and progression.
