摘要
目的 :通过中性粒细胞抗原 (NA)等位基因的频率调查 ,了解乌鲁木齐市维吾尔族人群 NA基因的多态性 ,为 NA相关的输血性疾病建立可靠的基因诊断技术。方法 :优化并建立了多重 PCR- SSP法 ,对 12 0名健康、无血缘关系的乌鲁木齐维吾尔族个体进行中性粒细胞抗原 (NA)的 NA1 和 NA2 种等位基因频率的调查。结果 :该样本中 NA1 基因频率为 0 .383;NA2 基因频率为 0 .6 17。结论:多重 PCR- SSP法分析 NA基因频率结果较为可靠 ,重复性较好。
Objective: To study the neutrophils antigen gene allele frequency of Uygur people in Urumqi region for gene diagnosis of some transfusion diseases. Methods: A optimum multiprimered PCR-SSP method was established to screen the netrophils antigen (NA) genotype of DNA samples isolated from 120 Uygur people blood donors in Urumqi region. Results: The allele frequency of NA-1 was 0.383 and NA-2 was 0.617 in this sample volume. Conclusion: The multiprimered PCR-SSP method is confirmed to be reliable and repeatable in our experiments for NA allele typing.
