脐血体积浓缩前存放的温度条件及时间界限 被引量:1

摘要 目的:探讨脐血采集后最佳的存放温度及时间期限。方法:采用集落培养及细胞活性计数方法研究不同条件下造血干祖细胞回收率。结果:采集后 4℃及室温存放其细胞活性差异无显著性 (P>0 .0 5 ) ,CFU- GM回收率差异有显著性 ,室温存放优于 4℃存放 (P<0 .0 5 ) ;存放时间上 ,造血干祖细胞集落形成能力的下降是一个渐进的过程 ,但 12 h以后集落产率开始下降更明显。 结论:脐血采集后 ,宜存放于室温并尽快进行脐血处理。 Objective: To study the temperature and time after collection of cord blood. Methods: Hematopoietic progenitor/stem cell culture and Trypan-blue detect cell viability. Results: Cell viability has′t significant difference on storage temperature (P=0.164); in the room temperature group, the recovery of CFU-GM higher than 4℃ group (P=0.043); the capacity of cell colony decreased gradually, decreased evidently 12 hours after collection. Conclusions: After cord blood collection, room temperature is fit for storage of cord blood, and cord blood should be manipulated as soon as quickly.
