摘要
目的 :建立同时测定血清中多种抗癫痫药物浓度的方法。方法 :血清用叔丁基甲醚 (MTBE)两次提取 ,室温下N2 吹干 ,残渣用甲醇溶解 ,进样分析 ,色谱条件为C1 8柱 ( 2 5 0mm× 4.6mm ,5 μm) ,流动相为 ψ(甲醇 ∶乙腈 ∶水 ) =40 ∶ 13∶ 47,检测波长 2 40nm。结果 :7种药物一次性分离 ,血清杂质不干扰测定 ,线性方程相关系数大于 0 .999,回收率为 92 %~ 10 5 %。结论 :方法专属性好 ,适用于临床血药浓度监测及药物的定性定量分析。
Objective:To establish a rapid and simple method for the determination of antipileptic drugs in serum simultaneously. Methods: The serum samples were extracted twice by MTBE. The residue was reconstituted in methanol and detected at 240 nm.The mobile phase consisted of methanol acetonitrile water (40∶13∶17 ).The YWC 18 (250 mm×4.6 mm,5 μm) column was used. Results: 7 drugs were well separated. The correlation coefficient of the linear equation was larger than 0.999. The recoveries were from 92% to 105%, and the variation coefficients of the between day and within day assay were less than 3%. Conclution: The method was simple,rapid and specific. It was suitable for qualitative and quantitative determination of toxic drugs in serum simultaneously.
