以香蕉 (Musaspp .)的横切薄层切片 (tTCL)外植体作为转化受体 ,研究影响根癌农杆菌转化香蕉的因素。研究表明 ,菌液的预处理和重悬液的pH值是影响香蕉转化的主要因子 ,而乙酰丁香酮 (AS)是香蕉遗传转化中必需的酚类物质 ,80 μmol/L是较理想的浓度 ;感染时间以 8~ 15min ,共培养的时间和温度分别以 4~ 5d及 2
In this stuby, transient GUS expression was used to optimize the biological conditons mediated by Agrobacterium tumefaciens for transverse thin cell layer (tTCL) of fruit Banana ( Musa spp.).Results showed that low generation of in vitro plantlets had greater shoot regeneration and higher transient GUS expression when tTCL was used. The pretreatment of Agrobacterium liquid,pH of resuspension (1/2MS) and the concentration of sugar were the main factors affecting banana genetic transformation,Acetosyringone (AS) (80 mo1/L) was the necessary phenol for banana genetic transformation other optimum conditions included 5 to 8 minutes of infection time, 4 to 5 days of co-cultivation at 26℃.
