摘要
利用高粱根作为实验材料 ,用不连续蔗糖密度梯度和DextranT - 5 0 0 /PEG - 335 0两相法探讨了纯化质膜的过程 ,以及去垢剂对K+运输蛋白的增溶效率。结果表明 ,由 6mL 4 0 % (W /W ) ,4mL 34% (W /W )和 4mL 2 2 % (W /W )蔗糖组成的不连续蔗糖梯度 ,经 80 0 0 0×g离心 3h后 ,在 34%~ 4 0 %界面处富含质膜。在质膜K+运输蛋白的增溶效果上 ,6
This paper described the procedure of membrane purification by step sucrose gradient and Dextran T 500/PEG 3350,and the solution efficiency of K + transport protein. The results showed that plasma membrane was mainly distributed between 34% to 40% sucrose solution,after centrifuged at 80,000×g for 3 h.In addition,6.5?mmol/L CHAPS was the best concentration for the solution of K + transport protein.
