摘要
目的 通过对原代培养乳鼠窦房结细胞进行光、电镜观察 ,了解培养窦房结细胞的形态结构特征。方法 取新生 2 4hWistar乳鼠的窦房结组织 ,用差速贴壁结合BrdU处理进行原代细胞纯化培养。结果 在培养的窦房结细胞中观察到有 3种不同形态的细胞 ,即梭形细胞、三角形细胞与不规则形细胞 ,其中梭形细胞最多 ,搏动频率最快 ,肌原纤维稀疏排列紊乱 ,细胞器不发达 ;三角形细胞与心房肌三角形细胞特征相似。结论 在培养的乳鼠窦房结细胞中 ,细胞体积小搏动频率快的梭形细胞即是窦房结的起搏细胞。
Objective To investigate the morphologic characters of primary cultured sinoatrial cells (SAN cells) under light microscope and transmission electron microscope with HE staining. Methods The SAN cells isolated from neonatal Wistar rat were cultured and purified with the method of differential attachment and BrdU-treatment. Results Three distinctly different types of cells were observed among the cultured cells of SAN, that is, spindle, triangle and irregular cells. The spindle cells covered the greatest proportion of the cultured cells of SAN. The frequency of spontaneous contraction of spindle cells was the highest among the contracting cells. The results of ultrastructure observation showed that myofibrils and other organelles in spindle cells were poorly organized and significantly decreased in number compared to triangle cells. There was no significant difference between triangle cells isolated from SAN and from atrial muscle. Conclusion Among the cultured cells from neonatal rat SAN, the spindle cells are the pacemaker cells of SAN.
