摘要
目的 研究大鼠骨髓基质细胞的体外培养及向心肌样细胞转化的条件。方法 取成年大鼠胫骨干骨髓基质细胞进行培养 ,保留附壁细胞传代。观察在培养液中添加诱导剂 5 -氮胞苷条件下骨髓基质细胞的生长和分化。结果 在形态上 ,大鼠骨髓基质细胞贴壁呈集落生长 ,有成纤维细胞样外观。 5 -氮胞苷诱导骨髓基质细胞转化为心肌样细胞 ,在诱导后 4周转化率接近 30 %。结论 骨髓基质细胞能够在体外被诱导分化为心肌样细胞 ,是自体心肌细胞一种良好的供体来源。
Objective To investigate the factors governing the growth and cardiomyogenic differentiation of rat marrow stromal cells (rMSCs) in vitro. Methods The rMSCs from adult rats, which attached to the walls in cell culture, were isolated and propagated for study. The cardiomyogenic induction was started by adding 5-azacytidine to the culture medium, and the proliferation and differentiation of them were observed. Results The rMSCs grew in colonies, toke the appearance of fibroblasts and differentiated into cardiac-like muscle cells after adding 5-azacytidine. The rate of differentiation approximated 30% in 4 weeks. Conclusion Cardiomyocytes can be generated from rMSCs in vitro,and the marrow stromal cell is one of the best resources of donor cardiomyocytes.
