摘要
目的 :建立一种适应性强、灵敏、准确的方法用于测定槐米中芦丁的含量。方法 :以 95 %乙醇为溶剂进行索氏提取 ;用醋酸乙酯 -甲酸 -水 (4∶ 1∶ 1)为展开剂分离 ;洗脱后以 10 % Al Cl3 甲醇溶液作为反应试剂 ,在 Ex=42 0 nm,Em=5 13 nm处测定荧光强度。结果 :此方法的线性范围为 0 .2~ 1.2μg/ml,平均回收率为 98.7% ,变异系数 RSD为 2 .71%。结论 :此方法稳定 ,重现性好 ,灵敏度高 。
OBJECTIVES: This paper reported a sensitive and accurate method for the assay of rutin in Flor Sophorae. METHODS: Rutin was extracted by 95% ethanol, and seperated from the others components with ethyl acetate formic acid water (4∶1∶1) as developer and eluted from silica gel G with methanol. 10% AlCl 3 methanol solution was added to react with rutin and the fluorescence of rutin was determined at Ex 420nm and Em 513nm. RESULTS: Linear range was between 0.2 μg/ml and 1.2μg/ml. The average recovery was 98.7%, with coefficient of variation of 2.71%. CONCLUSIONS: This method is feasible, stable, sensitive and has good repeatability, and could be used to control the quality of Flor Sophorae.
