The formation of rat osteoclast-like cell under regulation of different osteotropic hormones in vitro

摘要 目的 研究不同诱导因子对大鼠破骨细胞样细胞体外形成的影响。方法 建立大鼠脾细胞与成骨细胞的联合培养体系 ,将细胞悬液接种于预置盖玻片或牙本质片的 2 4孔培养板内 ,实验组分别加入骨吸收因子1,2 5 (OH ) 2 D3 、PTH、PGE2 ,对照组不加药 ,每 3d换培养液并加药 1次。结果 培养 5d后实验组可见多核细胞形成 ,并逐渐增多 ,TRAP染色阳性 ,电镜下见牙本质片上有吸收陷窝形成 ,其中以 1,2 5 (OH) 2 D3 组结果最明显。结论 体外培养大鼠破骨细胞样细胞的形成需要骨吸收因子的诱导 ,1,2 5 (OH) 2 D3 Objective To investigate the effect of different osteotropic hormones on the formation of rat osteoclast like cell in vitro.Methods We developed a co culture system with rat spleen cells and osteoblastic cells, and planted the cell suspension in 24 well tissue culture plate with cover slips or dentin slices. All cultures were fed every 3 days, different osteotropic hormones (1,25(OH) 2D 3, PTH, PGE 2 ) were added at the beginning of the culture and at each change of medium.Results After cultured for 5 days with osteotropic hormones, multinucleated cells were first observed, and their number increased gradually. They were postive in TRAP staining and could form absorption pit on dentin slices. The group of 1,25(OH) 2D 3 had the most significant results.Conclusion The formation of rat osteoclast like cell needs the regulation of osteotropic hormones, the effect of 1,25(OH) 2D 3 is most powerful.
