摘要
目的 观察扎鲁司特 (zafirlukast)对急性肺损伤的防治作用 .方法 新西兰大白兔 2 4只 ,随机分为对照组 (n=8) ,致伤组 (n=8) ,扎鲁司特治疗组 (n=8) .致伤组经颈静脉注入内毒素 (L PS,1m L· kg- 1 ) ,对照组经颈静脉注入生理盐水 (1ml· kg- 1 ) ,治疗组经颈静脉注入内毒素 (L PS,1m L· kg- 1 )前 1h及 30 min,胃内注入扎鲁司特 (2 0 mg· kg- 1 ) .测定各组肺动脉压、氧分压、外周血白细胞计数、肺泡灌洗液NO浓度、肺系数 (肺湿质量 /体质量 )及肺湿 /干比 ,并取肺活组织行 HE染色 .结果 治疗组肺动脉压及 NO浓度与致伤组无显著差别 (P >0 .0 5 ) ,氧分压较致伤组明显改善 (P <0 .0 1) ,肺系数和肺湿 /干比显著低于致伤组 (P<0 .0 1) ,外周血白细胞高于致伤组 (P<0 .0 1) .病理检查结果显示 ,治疗组肺损伤减轻 .
AIM To observe the preventive effect of zafirlukast against acute lung injury. METHODS 24 New-Zealand rabbits were randomly divided into three groups: Control group (n=8), injury group (n=8), and treatment group (n=8). into rats in injury group was Endotoxin (LPS, 1 mL·kg -1), normal saline (1 mL·kg -1) in control group, zafirlukast in treatment group (20 mg·kg -1) twice before being injected endotoxin (LPS, 1 mL·kg -1) half an hour and one hour. Observe the arterial oxygen pressure (PaO 2), the pulmonary arterial pressure, count of peripheral-blood leukocytes, NO concentration in the BALF, lung index (wet weight/body weight), lung's wet /dry weight ratio (W/D) and had HE staining of the pathological sections were observed respectively. RESULTS The pulmonary arterial pressure and NO concentration in treatment group had no significant difference with that in injury group (P>0.05), PaO 2 in treatment group significantly improved (P<0.01). Lung index and W/D ratio in treatment group were significantly lower than those in injury group (P<0.01), the count of peripheral-blood leukocytes in treatment group was higher than that in injury group (P<0.01). HE staining showed that lung injury in treatment group was improved. CONCLUSION Zafirlukast has preventive effect against acute lung injury.
