摘要
以不同器官为外植体,以不同浓度的外源生长素与细胞分裂素建立正交筛选实验,对诱导百子莲的愈伤组织进行了研究,结果表明:在百子莲愈伤组织诱导试验中,最佳外植体为花梗组织,其次为花丝组织;PIC对花梗外植体愈伤组织诱导具有明显的调控作用,诱导愈伤组织的最优配方是MS+2.0 mg/L PIC+30 g/L蔗糖;以百子莲花丝为外植体诱导愈伤组织的最优配方是MS+2.0 mg/L PIC+1.0 mg/L 6-BA+30 g/L蔗糖。
This research investigates the callus of Agapanthus praecox ssp.Orientalis by taking different organs as explant and utilizing different concentrations of exogenous auxin and cytokinins to establish the orthogonal screening experiment.The experimental result indicates that the optimal explant is pedicel tissue,and filaments organization comes second in the experiment of callus tissue inducement of agapanthus ;PIC plays an apparent regulating role in the callus induction.The optimum formula for inducing callus is MS + 2.0 mg/L PIC + 30 g/L sugar cane,and the optimum formula for using agipanthus filament as the explant to induce callus is MS + 2.0 mg/L PIC + 1.0 mg/L 6-BA + 30 g/ L sucrose.
