摘要
本试验研究了二噁英(Dioxin)对大鼠机体的损伤及自由基清除剂(free radical scavenger,FRS)的保护效应。动物随机分为3组:对照组(Ⅰ组)、Dioxin组(Ⅱ组)和Dioxin+FRS保护组(Ⅲ组)。3组动物经相应处理后分别在4、8、12、16 d采集血液和肝脏作为检测样本。试验结果显示:Ⅱ组丙二醛(malondialdehyde,MDA)水平在4、8、12、16 d测定时明显高于Ⅰ(P<0.01)、Ⅲ组(P>0.05),Ⅱ组超氧化物歧化酶(super oxide dismutase,SOD)水平在4、8、12、16 d测定时明显高于Ⅰ(P<0.01)、Ⅲ组(P>0.05)。病理组织学变化:Ⅱ组主要表现为空泡变性,超微结构变化表现为线粒体肿大、线粒体嵴减少、溶解,出现异形线粒体;细胞核形态改变,出现不规则形状,染色质成块边集于核膜层;滑面内质网池扩张、融合。Ⅲ组出现类似变化,但程度较轻;Ⅰ组未见异常变化。研究表明,Dioxin导致大鼠机体严重的损伤,FRS对其具有良好的颉颃作用。Dioxin急性中毒可导致肝脏损伤,机体内自由基水平升高,从而导致机体的损伤;而FRS则能明显抑制Dioxin对肝脏的损伤,从而抑制各种毒性效应,对由Dioxin诱导的肝损伤具有显著的保护效应。进一步说明了体内自由基(free radical,FR)水平的升高在Dioxin中毒的发生、发展中发挥了重要作用。
The effect of Dioxin on organism of rats and the protective effect of FRS were studied in this paper.Seventy-two rats were randomly divided into three groups:the controls group (group Ⅰ),Dioxin treatment group (group Ⅱ) and FRS and Dioxin treatment group (group Ⅲ).FRS was used to study the antagonistic effects of Dioxin.Blood and liver samples were taken after three,four,eight,twelve and eighteen days.Compared with control group,the SOD in the Dioxin group significantly decreased (P 〈 0.01) and the MDA levels significantly increased (P 〈 0.01).The pathological lesions of liver were characterized by vacuolar degeneration.The ultrastructural changes were marked with the swelling of mitochondrial with destruction of crista and heterotypic mitochondrial,dilation of the cisternae and vesicles-like proliferation of smooth endoplasmie reticulum,and marginal immigration of nucleolus and chromatin.In group Ⅱ,the ultrastructural changes were marked with mitochondrial crista destruction and solution,but these were no abnormal changes in normal control group (group Ⅰ).The results showed that rat Dioxin poisoning could cause severe damages of liver,and educe various kinds poisoning effects.The elevation of free radical level played great important role in histopathological damages,the development of Dioxin poisoning in rats.FRS had significant protective effect on injuries of Dioxin in the liver.
