守宫硫酸糖肽对人乳腺癌细胞SKBR3增殖的影响及其机制 被引量:2

摘要 目的观察守宫硫酸糖肽(GSPP)对人乳腺癌细胞SKBR3增殖的影响,并探讨其作用机制。方法体外培养SKBR3细胞,分别加入GSPP 100μg/mL、碱性成纤维细胞生长因子(bFGF)5 ng/mL、GSPP 100μg/mL联合bFGF 5 ng/mL,并设PBS阴性对照组。72 h后分别用台盼蓝染色法检测增殖细胞数,流式细胞仪检测细胞周期,ELISA法检测CyclinD1。结果与阴性对照组比较,GSPP对SKBR3细胞增殖有抑制作用(P<0.01),bFGF对SKBR3细胞增殖有促进作用(P<0.01),二者合用细胞数目与PBS阴性对照比较无统计学差异。GSPP将细胞周期阻滞在G0/G1期,并抑制CyclinD1分泌(P均<0.05);bFGF降低G0/G1期细胞比例,并促进CyclinD1分泌(P均<0.05);GSPP同bFGF合用的G0/G1期细胞比例及CyclinD1分泌均与PBS阴性对照组比较无统计学差异。结论GSPP通过抑制CyclinD1分泌抑制SKBR3细胞增殖;GSPP可拮抗bFGF对细胞增殖的促进作用。 Objective To observe the effects of Gekko sulfated glycopeptides ( GSPP ) on the proliferation of human breast cancer SKBR3 cells and to explore its mechanism .Methods Human breast cancer SKBR3 cells were cultured in vitro.GSPP 100 μg/mL, basic fibroblast growth factor ( bFGF) 5 ng/mL, GSPP 100 μg/mL and bFGF 5 ng/mL were added to SKBR3 cells, respectively.Moreover, PBS negative control group was designed in the study .After 72 h, cell pro-liferation was detected by trypan blue staining method;cell cycle was tested by flow cytometry;and the secretion of cyclin D1 was tested by ELISA .Results Compared with the negative control group , GSPP had inhibitory effect on cell prolifera-tion of SKBR3 cells (P&lt;0.01), and bFGF had promoting effect on cell proliferation (P&lt;0.01).The number of cells in the combination group had no difference between the combination group treated by GSPP and bFGF and the PBS negative control group.The GSPP arrested cells in the G1 phase and decreased the secretion of cyclin D1 (all P&lt;0.05).The bFGF decreased the proportion of cells in G 0/G1 phase and increased the secretion of cyclin D 1 (all P&lt;0.05).The proportion of cells in the G0/G1 phase and secretion of cyclin D1 in the combination group had no statistical difference as compared with that of the PBS negative control group .Conclusions GSPP can inhibit SKBR3 cells proliferation by decreasing the level of cyclin D1.What&#39;s more, GSPP acts as an antagonist for bFGF and inhibits the promoting effect of bFGF on cell prolifera-tion.
