吗啡对培养的尾核神经元电压门控性钙通道的影响 被引量:5

摘要 目的 探讨尾核神经元在吗啡镇痛中的离子机制。方法 采用膜片钳全细胞记录模式观察了 2 2例尾核神经元电压门控性钙通道在吗啡给药前后的电流变化。结果 观测的 2 2例细胞中 ,9例 (40 9% )细胞加入吗啡后 ,ICa由 1 1 85 .43pA± 1 84 2pA增加到 1 4 2 4 .8pA± 2 59.5pA ,与给药前相比增加了 2 0 .2 1 % ,其差异经配对t检验 ,具有显著性意义 (P <0 .0 5)。 7例 (31 8% )细胞加入吗啡后 ,ICa由 1 350 3pA± 2 0 3 .7pA减少到 1 0 50 6pA± 1 79.6pA ,降低 2 2 .2 % ,其差异具有显著性意义 (P <0 .0 5)。吗啡作用可被纳洛酮翻转。 6例细胞 (2 7.2 % )未观察到明显改变。结论 吗啡不仅可在整体情况下 ,经不同核团神经元之间的联系对尾核神经元的电活动进行调节 ,同时 。 Objective To investigate the ion mechanism of caudate nucleus neurons underlying morphine ease pain process.Methods Whole cell patch clamp technique was used to study the changes of voltage dependent calcium current of 22 neurons in caudate nucleus before and after morphine treatment.Results Voltage dependent calcium current was increased after morphine treatment in 9 of the 22 neurons.It changed from 1185.43pA±184.2pA to 1424.8pA±259.5pA,the difference had marked significance( P <0.05).I Ca decreased after adding morphine in 7 of the neurons from 1350.3pA±203.7pA to 1050.6pA±179.6pA which has marked significance too( P <0.05).The effects of morphine could be overturned by naloxone.No distinct changes was found in 6 neurons.Conclusion Morphine may adjust channel activity of neurons in caudate nucleus not only by contacts of different nucleus,but also by acting on single neuron in caudate nucleus directly.
