摘要
目的 研究 β2 -肾上腺素能受体 (β2 - adrenoceptor,β2 - AR)基因编码区 +10 5 3和 +12 39位单核苷酸多态性 (single nucleotide polymorphisms,SNPs)与老年高血压的关系。方法 采用聚合酶链反应 -限制性内切酶片段长度多态性技术 ,检测 86例高血压患者 ,43名健康对照者的 β2 - AR基因 +10 5 3位和+12 39位 SNP基因型。结果 两种 SNPs基因型频率在正常人群分布符合 Hardy- Weinberg平衡。 +12 39位 SNP的基因型频率和基因频率在老年中、重度高血压组和对照组分布差异有显著性 (P<0 .0 5 ) ;+10 5 3位 SNP基因型频率和基因频率在两组人群中分布差异无显著性 (P>0 .0 5 )。结论 β2 - AR基因 +12 39位SNP可能与老年高血压相关 ,+10 5 3位 SNP仅为基因多态性标志。
Objective To investigate the association of two single nucleotide polymorphisms (SNPs) of β 2-adrenoceptor (β 2-AR) gene with hypertension in elderly patients.Methods The study samples were collected from unrelated Chinese Han population of Dabie Mountain in Anhui province. Eighty-six elderly patients with hypertension and 43 controls were selected. Genotypes of +1053 and +1239 SNPs were typed by polymerase chain reaction-restriction fragment length polymorphism.Results The frequencies of the two SNPs complied well with the Hardy-Weinberg equilibrium in normal group. The distribution of genotypes AA, GA,GG of the SNP at locus+1239 in moderate and severe hypertension group was significantly different from that in normal group (χ 2=8.67, P <0.05). There were evident differences in the frequencies of alleles of the two groups (χ 2=4.02, P <0.05). No significant difference was observed in the distribution of genotypes of the SNP at locus+1053 between the two groups.Conclusion These data indicate that the SNP at locus+1239 of β 2-AR gene is associated with hypertension in elderly patients.
