抗病毒新途径:TFO切割病毒DNA研究进展

摘要 慢性病毒感染性疾病的治疗仍是一个临床难题。近年来对三股螺旋形成寡核苷酸(TFO)的研究表明,TFO与双链DNA结合成三链DNA具有高度的序列特异性,TFO末端经金属配合物(啡咯啉铜、卟啉锰、甲磺酸去铁胺铜、乙二胺四乙酸铁)、细胞毒药物(博来霉素、玫瑰树碱)、同位素(125I、111In)光敏物质(菲并二氢二恶英、恶唑黄、氨基-P-喹吖啶)、酶激活剂(吲哚并咔唑)等修饰,能显著提高TFO对靶 DNA的亲和性和切割能力。目前已对多种病毒核酸分子内三股螺旋结构的形成进行了研究,并有学者试用卟啉锰、氨基-P-喹吖啶化合物修饰的TFO定点切割HIV基因中的靶序列,在体外实验中获得了可喜的成效。但TFO在临床的应用还有许多问题需进一步研究。
