胃肠道恶性肿瘤端粒酶RNA表达及其与p53蛋白的相关性研究

摘要 目的 :探讨胃肠道恶性肿瘤中端粒酶RNA(hTR)表达意义及其与肿瘤抑制基因p5 3的相关性。方法 :采用Lightcycler实时逆转录荧光定量PCR方法定量检测胃肠道恶性肿瘤中hTR的表达 ;用免疫组织化学技术检测p5 3蛋白 ,比较hTR与p5 3表达间的关系。结果 :胃肠道恶性肿瘤组与正常组的配对t检验显示hTR的表达差异有显著性 (P <0 .0 5 )。p5 3阳性与p5 3阴性胃肠道恶性肿瘤组中hTR的表达差异无显著性 (P >0 .0 5 )。结论 :胃肠道恶性肿瘤组织与正常组织的hTR表达有差异 ,表明定量检测hTR表达有助于胃肠道恶性肿瘤的早期诊断。hTR表达不直接受p5 3基因变化的影响 ,两者在胃肠道恶性肿瘤发生中存在各自不同的调控途径。 Objective:To explore the expression of human telomerase RNA (hTR) and its relationship to tumor suppressor gene p53 in gastroenteric carcinomas. Methods:A quantitative real-time RT-PCR assay was applied to quantify the expression of hTR and immunohistochemistry (IHC) was used to observe the p53 protein expression in 18 specimens of gastroenteric carcinomas and their corresponding non-cancerous tissues. Both the significance of hTR expression in and its relationship with p53 protein were investigated. Results: There is significant difference of hTR expression between the gastroenteric carcinomas group and normal tissue group( P <0.05).In the difference of hTR expression,there is no statistic difference between p53 negative group and p53 positive group ( P >0.05). Conclusions: There is difference of hTR expression between gastroenteric carcinomas and normal tissues. hTR expression detected by the quantitative real-time RT-PCR assay will be helpful to early diagnosis of gastroenteric carcinomas. hTR expression is independent of the p53 protein expression.
