摘要
目的 :探讨转录因子NF κB对砷制剂诱导下NB4细胞程序性死亡的调控作用。 方法 :将NB4细胞与不同浓度的三氧化二砷共同孵育 ,在不同时间用流式细胞仪检测胞质内转录因子NF κB的表达 ,同时作细胞周期分析、PI和HO双染以及DNA梯度 ,以鉴定As2 O3的致程序性死亡的作用。 结果 :NB4细胞在 1μmol/L、2 μmol/LAs2 O3的诱导下 ,在 16~ 2 4h的期间内均有明显的程序性死亡现象。NF κB在 6~ 16h期间的对照组和加药组 ( 1μmol/L的As2 O3)均有表达 ,对照组中 12h后表达明显下降 (P <0 .0 5 ) ,加药组较对照组也明显减低 (P <0 .0 5 ) ,但不存在时间依从性。 结论 :As2 O3可诱导早幼粒细胞程序性死亡 ,在这一过程中 ,NF
Objectives:To explore the effect of transcription factor NF κB on apoptosis of NB 4 cell line induced by arsenic trioxide. Methods:The NB 4 cells were cultured in RPMI 1640 with different concentration of As 2 O 3 .These cells were harvested at different cultured time. Meanwhile, the expression of NF κB was examined by flow cytometry . In addition, the apoptosis induced by As 2 O 3 was tested with the analysis of cell cycle, DNA ladder, Propidium Iodide and Hoechst 33342 stain. Results:The cells of two groups expressed NF κB after cultured 6~16 hours. The expression of NF κB reduced in As 2 O 3 group without time dependent pattern. To our surprise ,the expression of NF κB of the cells in the control group was remarkably decreased after 12 hours of culture( P <0.05). Conclusions: As 2 O 3 can induce NB 4 cells apoptosis. It might be the main machanism of therapy of As 2 O 3 in APL. Expression of NF κB in NB 4 cells induced by As 2 O 3 was decreased. It suggested that NF κB may be a potential apoptotic suppressor.
