E-钙黏蛋白复合体与胃癌 被引量：5

E-Cadberin complex and gastric cancer

摘要 E-钙黏蛋白是一类介导细胞-细胞间相互黏附、具有维持组织结构完整性和极性的钙依赖性跨膜蛋白。E-钙黏蛋白功能的发挥,有赖于与连接素共同构成复合体。研究发现,上皮型恶性肿瘤的分化、侵袭和转移与E-钙黏蛋白-连接素复合体的表达及功能障碍有关。众多研究表明,E-钙黏蛋白-连接素复合体介导的黏附功能下降可能存在三个机制:E-钙黏蛋白表达下调或基因突变;α-连接素表达异常或基因缺失;β-连接素分子异常生化修饰。因β-连接素是APC-β-catenin-Tcf/Lef信号转导通路的关键分子,E-钙黏蛋白-连接素复合体介导的黏附功能下降还可能涉及癌细胞生物学中更深、更广的层面。对E-钙黏蛋白-连接素复合体的深入研究将有助于进一步揭示胃癌等恶性肿瘤的发生、发展机制。
