老年性骨质疏松患者松质骨中骨形成蛋白-2及骨形成蛋白-7基因表达

Study of the gene expression of bone morphogenetic protein(BMP)-2,-7 in the cancellous bone of senior primary osteoporosis

目的通过观察老年性骨质疏松患者与正常人松质骨中骨形成蛋白(BMP)2、7mRNA含量的变化,探讨BMP基因表达在老年性骨质疏松症发病机制中的作用。方法共选8例临床病例,根据患者X线表现、桡骨远端骨密度值以及生化指标检测确定为老年性骨质疏松患者。对照组取8例年龄在20～30岁之间的骤然死亡的健康男性股骨头,取各自的股骨头提取RNA,以BMP-2、7cDNA为探针进行RNA斑点杂交,根据图像分析结果进行统计处理,比较两组差异。结果临床患病组BMP-2、BMP-7mRNA含量明显低于对照组。结论老年性骨质疏松症患者松质骨中BMP-2及BMP-7mRNA表达明显降低,这可能是老年性骨质疏松症发病原因之一。

Objective To explore the role of gene expression of BMP in the pathogenesis of senior osteoporosis by observing changes in the content of BMP 2 mRNA and BMP 7 mRNA in the cancellous bone of patients with senior osteoporosis.Methods We chose 8 patients as the experiment group according to their X ray photography, BMD in distal end of radius and biochemical tests.The control group was composed of 8 healthy males between 20 and 30 years old. We extracted total RNA from each one's cancellous bone in the femur head and made dot blot hybridization by using BMP 2,7cDNA as probes , then compared the results by imaging analysis.Results The content of both BMP 2 and BMP 7 was lower in the group of patients with senior osteoporosis compared with control group.Conclusion The low expression of BMP 2 and BMP 7 mRNA in the cancellous bone of patients with senior osteoporosis might be one of the pathogeny of senior osteoporosis.
