摘要 目的 用rhGM CSF联合rhIL 4将慢性粒细胞白血病 (CML)原代细胞诱导成树突状细胞 (DC) ,探索白血病免疫治疗的新途径。方法 分离初诊CML患者外周血单个核细胞 (PBMNC) ,用rhGM CSF联合rhIL 4共同培养 7d ;流式细胞仪 (FACS)检测细胞因子诱导前后细胞表面HLA DR、CD1a、CD80和CD86的表达 ,电镜观察细胞形态。结果 经 7d的诱导 ,HLA DR、CD1a、CD80和CD86的表达明显上调 ;电镜观察到典型的DC形态特征。结论 rhGM CSF联合rhIL 4可成功将CML原代细胞诱导成CML DC 。 Objective To induce dendritic cells (DCs) from primary chronic myeloid leukemia(CML) cells with rhGM CSF and rhIL 4, so as to provide a new approach for immunotherapy of leukemia. Methods MNCs were isolated from the peripheral blood of CML patients when diagnosed,then co cultured with rhGM CSF and rhIL 4 for 7 days.Before and after the induction by the cytokines,HLA DR、CD1a、CD80 and CD86 expression were assayed by flow cytometry,the morphologic features of DC were observed by electronmicroscope. Results After cultured for 7 days, the expressions of these phenotypes were upregulated significantly. Typical morphologic characteristics of DC were observed.Conclusion Primary CML cells can give rise to CML DC by rhGM CSF and rhIL 4, suggesting that it may be used efficiently in immunotherapy of CML.
