62例恶性肿瘤尿多胺类化合物固相萃取高效液相色谱法检测价值的研究 被引量:3

摘要 目的 探讨人尿多胺改良高效液相色谱测定方法及其对恶性肿瘤患者的应用价值。方法  (1)分析常规抗癌药物如 ,5 氟脲嘧啶 (5 Fu)、长春新碱 (VCR)、碳铂 (Carboplatin)、顺铂 (CDDP)、足叶乙甙 (VP16 )、放线菌素D(Act.D)、丝裂霉素C(MMC)等对固相萃取 高效液相色谱 (solidphaseextraction highperformanceliquidchromatography ,SPE HPLC)技术测定多胺的干扰 ;(2 )采用SPE HPLC ,测定 6 2例恶性肿瘤患者 (36例可手术和 2 8例不能手术之晚期肿瘤患者 )及 2 8例对照健康者尿多胺水平 ;(3)对比检查16例晚期肿瘤化疗患者及 16例化疗中 (初始阶段 )的患者和 2 6例晚期肿瘤化疗前对照者的尿多胺。结果 可手术切除组与不能手术之晚期肿瘤组恶性肿瘤患者尿总多胺、Pu、Sm及Sd水平和Sd/Sm、Pu/Sm比值明显高于健康对照组 (P <0 0 5或 <0 0 1) ;晚期肿瘤化疗一疗程后尿总PAs、Pu、Sd及Sm水平均降低 (P <0 0 5 ) ,而化疗初始尿总PAs、Pu、Sd/Sm及Pu/Sm比值呈一过性增高 (P<0 0 5或P <0 0 1)。结论 采用固相萃取 高效液相色谱分析法 ,用于恶性肿瘤患者尿多胺检测结果表明 :恶性肿瘤患者尿多胺在肿瘤细胞生长中起重要作用 ,尿液多胺作为生物标志物 ,其SPE HPLC法检测对于临床恶性肿瘤患者诊断与化疗效果的? Objective To introduce an improved SPE HPLC assay for the quantitative analysis of urine polyamines in different kinds of cancers.Methods (1)The interference of several clinical anticancer agents(5 Fu, VCR, Carboplatin, CDDP,Act.D,MMC, et al) in the determination of polyamines was studied.(2)The levels of the urine PAs were detected using SPE HPLC in 36 operable patients with cancers and 26 inoperable patients.28 healthy persons served as normal control.(3)The levels of the urine PAs were examined using SPE HPLC in 16 advanced cancer patients after chemotherapy and 16 patients in the beginning of chemotherapy, as well as in 26 patients used as controls.Results The levels of the urine PAs, Pu, Sd and Sd/Sm as well as Pu/Sm values were higher in the operable cancer patients and much higher in the inoperable patients than in healthy persons( P <0 05 or P <0 01);The levels of the urine PAs, Pu, Sd and Sm in the advanced cancer patients after a course of chemotherapy, were lower than prechemotherapy ( P <0 05);However, the levels of the urine PAs, Pu and Sd/Sm as well as Pu/Sm values of the patients with advanced cancer in the beginning of chemotherapy, were higher than in the control group( P <0 05 or P <0 01).Conclusion The clinical tests of polyamines for the patients with advanced cancer using SPE HPLC, suggest that urine PAs are a crucial factor in cancers and play an important role in cancer cell growth.The urine PAs as biochemical marker, are valuable in helping to diagnose cancers and evaluate the curative effects of chemotherapy.
