人重组生长激素对豚鼠离体心脏血流动力学及心肌细胞动作电位影响 被引量:2

Effects of human recombinant growth hormone on haemodynamics of isolated guinea-pig heart and action potential in isolated guinea-pig ventricular myocytes

摘要 目的 :观察人重组生长激素对正常豚鼠强心作用和对心室肌细胞动作电位的影响。方法 :采用langendorff离体心脏灌流法 ,悬浮玻璃电极法。结果 :注入人重组生长激素 10min后 ,左室最大收缩压 ,左室舒张末压 ,左室内压的最大升降速率的差值分别为 (6 .7± 5 .5 )kPa ,(0 .5 0± 0 .10 )kPa ,(139± 93)ps·s- 1,明显升高 ,心率差值为 (10± 16 )次·min- 1减慢 ,动作电位振幅值 (APA) ,超射 (OS) ,复极 5 0 %及 90 %水平的动作电位时程 (APD50 ,APD90 )分别为 (13.2± 6 )mv ,(9.6± 2 .2 )mv ,(84±2 9)ms,(90± 2 2 )ms ,显著延长 (均P <0 .0 1)。结论 :人重组生长激素对正常豚鼠具有正性肌力作用 。 AIM: To investigate the positive inotropic effects of human recombinant growth hormone (hr-GH) on the normal guinea-pig's heart and the influence of hr-GH on the action potential of the ventricular myocytes in isolated guinea-pig's heart. METHODS: Both methods of the langendorff's isolated heart perfusion and the glass suspension electrode were used. RESULTS: Ten minutes after the hr-GH administration for the experimental model, left ventricular systolic pressure, ±dp/dt max and left ventricular end-diastolic pressure increased obviously while heart rate decreased. The overshoot action potential duration (APD 50,APD 90), action potentialamplitude (APA) of action potential were incerased prominently. There were obvious statastic difference between the records before and after hr-GH perfusion. CONCLUSION: The hr-GH shows the positive inotropic effect on the isolated guinea-pig heart. The probable mechenism may come from the increasing of the Ca 2+ amount in myocytes.
