摘要
目的 制备HLA -B2 7单克隆抗体。方法 用HLA -B2 7抗原免疫小鼠 ,取脾细胞与骨髓瘤细胞融合制备杂交瘤细胞 ,用ELISA法筛选分泌该单抗的细胞株。结果 建立了分泌该单抗的细胞系 3B3和 4F2 ,抗体均为IgG2b ,3B3细胞分泌的单抗只与B2 7反应 ,而 4F2细胞分泌的单抗与B7有交叉反应。 结论 建立了高特异性分泌抗B2 7单抗的细胞株 。
Objective To preparation HLA-B27 monoclonal antibody.Methods HLA-B27mAb was derived from fusion of mouse SP2/0 myeloma cells with spleen cells from BALB/c mouse immunized with a purified HLA-B27 antigen using a conventional fusion technique. ELISA method was used for screening.Results Two clones named 3B3 and 4F2, belonging to IgG2b , were finally obtained. The specificity of these mAbs was examined by microcytotoxicity test. No cross reactivity with B7 and B22 antigens was detected in 3B3 cell lines, but there was cross reactivity between B27 mAb(from 4F2cell lines) and B7 antigen.Conclusion HLA-B27 mAb (3B3) was a highly specific monoclonal antibody.
