摘要
目的 :检测肺癌组织中黑色素瘤抗原 3基因 (MAGE 3)mRNA的表达。方法 :用逆转录 套式聚合酶链反应 (RT PCR)对 31例肺癌患者癌组织和相应癌旁组织MAGE 3mRNA表达情况进行测定 ;PE 377DNA测序仪对 5例10个RT PCR扩增产物中的目的基因片段进行DNA序列测定。结果 :31例肺癌患者癌组织中 2 6例表达MAGE 3mRNA ,阳性率为 83.9% ;相应的癌旁组织均未表达。DNA序列测定证明PCR扩增产物中目的基因片段均为MAGE 3cDNA序列 ,所测 5例样本中 4例样本有 2个相同位置的碱基发生了点突变 ,导致一个氨基酸残基改变。结论 :①MAGE 3mRNA在肺癌中呈高比例表达 ,提示此抗原有可能作为肺癌患者免疫治疗的靶抗原 ;②我国肺癌患者中存在MAGE
Aim:To investigate the expression of MAGE 3 mRNA in tissues derived from lung cancer and to discuss the possibility of using MAGE 3 antigen as a new peptide vaccine for immunotherapy for cases of lung cancer. Methods: The tissues of lung cancer and the adjacent tissues of the lung were obtained from 31 cases of lung cancer. Total RNA was extracted and cDNA was synthesized. Nested Polymerase Chain Reaction using MAGE 3 specific primer was performed to detect the expression of MAGE 3.The 10 clones of 5 cases, MAGE 3 mRNA positive PCR products, were sequenced by using PE 377 DNA sequencer. Results: Of 31 cases of lung cancers, 26 (83.9%) expressed MAGE 3 mRNA. The expression of MAGE 3 gene was not detected in adjacent tissues. The DNA sequence test confirmed that the target gene fragment in all 5 PCR products was MAGE 3 cDNA. Point mutations occurred in 4 samples(8 clones) as a result of alternation of amino acid residue in one position. Conclusions: ①The MAGE 3 gene was expressed exclusively with a high positivity rate in tumor tissues from cases of lung cancer. This tumor rejection antigen may have potential use as a new peptide vaccine for immunotherapy for lung cancers. ②There are two point mutations of MAGE 3 gene sequence in some Chinese cases of lung cancer.
