摘要
目的 :观察外源性磷酸肌酸对大鼠供心保存效果及其保护机制。方法 :4 0只健康雄性SD大鼠随机均分A、B 2组 ,移植前供心保存 2 4 0min ,A组供心用 4℃St.Thomas液保存。B组保存液中添加外源性磷酸肌酸。保存后进行腹腔异位心脏移植术 ,并观察供心复跳情况。 2 4h后处死受体动物 ,切除移植心脏 ,测心肌组织中脂质过氧化物 (LPO)、超氧化物歧化酶 (SOD)、三磷酸腺苷 (ATP)含量 ,电镜观察心肌超微结构改变。结果 :B组移植心脏自动复跳率高于A组 ,心律失常发生率低于A组。B组心肌组织中SOD、ATP含量均高于A组 (P <0 .0 5 ) ,LPO含量低于A组 (P <0 .0 1) ,心肌超微结构损伤较A组轻。结论 :外源性磷酸肌酸能明显增强保存液对心脏保护作用 。
Aim:To study the effects of creatine phosphate (CP) on donor heart preservation for rats.Methods:Forty rats were randomly allocated into 2 groups.The donor hearts were preserved with cardioplegia for group A,and cardioplegia plus PC for group B.The donor hearts were transplanted into the abdomen of recipient rats.The left ventricular tissues were obtained from the donor hearts after 24 h of transplantation.The contents of SOD,LPO,and ATP were determined and cardium ultrastructure were observed.Results: The contents of SOD and ATP were significantly higher in the group B than that in group A. The contents of LPO were significantly lower in group B than that in group A.The incidence of arrhythmia in group B was much lower than that in group A. Conclusion: Exgenous creatine phosphate could significantly strengthen cardioplegia effects.
