肺纤维化大鼠丙二醛、肿瘤坏死因子-α和内皮素-1水平的变化

Changes of malondialdehyde,tumor necrosis factor-α and endothelin-1 in pulmonary fibrosis in rats

目的 :探讨平阳霉素致大鼠肺纤维化的机制。方法 :2 0只大鼠随机等分为模型组和对照组 ,模型组气管内滴入平阳霉素建立肺纤维化模型 ,对照组滴入生理盐水 ,分别于滴药后第 7d ,2 8d ,每组随机处死 5只取样 ,观察肺组织中丙二醛 (MDA)、内皮素 1(ET 1)、羟脯氨酸 (HYP)和支气管肺泡灌洗液 (BALF)中白细胞总数和肿瘤坏死因子 α(TNF α)的水平变化。结果 :第 7d模型组BALF中白细胞总数、TNF α和肺组织匀浆MDA、ET 1的含量显著高于对照组 (P均 <0 .0 5 ) ;第 2 8d白细胞总数和ET 1模型组仍高于对照组 (P均 <0 .0 5 ) ;MDA和白细胞总数、TNF α呈正相关 (r分别为 0 .83和 0 .6 8,P均 <0 .0 0 1) ;TNF α和白细胞总数、ET 1亦呈正相关 (r分别为 0 .6 7和 0 .6 6 ,P均 <0 .0 1)。结论 :MDA、TNF α和ET

Aim:To explore the mechanism of the pulmonary fibrosis induced by pingyangmycin in rats.Methods:Twenty rats were randomly allocated into experiment group and control group. Each group had 10 rats.Pingyangmycin and normal saline was intratracheally given to the experiment group and control group,respectively.Samples were collected on the 7th and 28th d after intratracheal adminitration. Five rats were selected randomly from each group each time.The levels of malondialhyde, endothelin 1,and HYP in the lung tissues,the total number of leucocyte,and the level of tumor necrosis factor α(TNF α) in the bronchoalveolar lavage fluid(BALF) were measured.Results:On the 7th d,the total number of leucocyte, the levels of TNF α,MDA,ET 1 and HYP were significantly higher in the experiment group than those in the control group( P <0.05).On the 28th d,the total number of leucocyte and the levels of ET 1 and HYP were still higher in the experiment group(both P <0.05).The level of MDA was positively correlated with both the total number of leucocyte and the level of TNF α ( r =0.83 and 0.68 respectively,both P < 0.001 ); the level of TNF α was also positively correlated with the total number of leucocyte and the level of ET 1( r =0.67 and 0.66 respectively,both P < 0.01 ).Conclusion: MDA,TNF α, and ET 1 all participate in the mechanism of pulmonary fibrosis induced by pingyangmycin.
