摘要
在生物转化法生产麻黄碱研究过程中 ,建立了一种检测转化液中麻黄碱含量的方法———薄板层析双波长扫描法 .该方法以硅胶GF2 54 为吸附剂 ,以甲醇 浓氨水混合液 (V(甲醇 )∶V (浓氨水 ) =98∶2 )为展开剂 ,0 .2 %茚三酮丙酮溶液为显色剂 ,扫描波长λS=5 10nm ,λR=70 0nm .并运用该法考察了 4株初筛菌株的转化能力 .
A method (dual wave length TLC scanning method) was established to determine ephedrine in biotransformatic mixture during the study on the production of ephedrine by biotransformation. Ephedrine was determined on silica gel GF 254 plate with methanol ammonia water(98∶2) as developing solvent. Color developing reagent was 0.2% ninhydrin (acetone solution), and wave lengthes for scanning were λ S=510 nm,λ R=700 nm. The biotransformatic ability of four preliminary screened strains was detected with this method.
