高剪切速率诱发血小板激活作用的研究

摘要 血小板α-颗粒膜蛋白((GMP-140)在血小板膜表面的暴露是血小板被激活的重要标志。用抗GMP-140单克隆抗体做探针,以8条杂种犬为对象,研究了高剪切速率(230s^(-1),5min)作用于全血后血小板功能的变化。用放免测定方法测得高剪切速率作用后血小板膜GMP-140克分子数量显著增多(P<0.01),间接免疫荧光结合计算机图像分析提示染上荧光血小板数及其占有的总面积均显著增加(P<0.01);阻抗法和电镜观察到血小板聚集也相应增加(P<0.01)。结果提示:高剪切速率可激活血小板并使其很快聚集,这可能是血管狭窄导致局部高剪切速率,致血栓形成的重要原因。 In order to study activation of platelet induced by high shear rate (HSR), 8 healthy mongrel dogs weighting 14-17 kg were used for experimental tests. Results:(l)moles of GMP-140/platelet after HSR being much elevated than the control group (P>0.01). (2)Ω value of platelet aggregation induced by adrenaline being much decreased after HSR (P<0.01). (3) number of fluorescent platelet and their total occupied area being markedly increased (P <0.01).(4)The pile of platelet aggregation being also increased under the electron microscope. Our data suggested that HSR could increase the activity of platelet and enhance the aggregation of platelet.
