摘要
用RAPD PCR技术初步研究了中国野桑蚕与日本福冈野桑蚕之间的DNA多态性。结果表明 ,不同地域中国野桑蚕个体之间及其与日本福冈野桑蚕个体之间均呈现出丰富的DNA多态性。中国野桑蚕同家蚕共有带率达 3 5 5 % ,同日本福冈野桑蚕为 4 5 1% ,而日本福冈野桑蚕同家蚕为 3 2 % ;中国野桑蚕与家蚕的平均遗传距离为0 5 2 ,日本野桑蚕与家蚕的平均遗传距离为 0 6 3,与中国野桑蚕的为 0 5 8,而且与中国沈阳地区野桑蚕之间的遗传距离最近 ,为 0 4 8,以此创建了它们的系统进化树。
Preliminary RAPD PCR study was conducted on the polymorphism of Bombyx mandarina M.and Japanese (Fukuoka) Bombyx mandarina M.Results showed that there exists rich DNA polymorphism between individuals of both Bombyx mandarina M. and Japanese (Fukuoka) Bombyx mandarina M.The rate of shared bands between Bombyx mandarina M.and Bombyx mori was 3.55%,while it was 3.2% between Japanese(Fukuoka) Bombyx mandarina M.and Bombyx mori .The mean genetic distances between Bombyx mandarina M.and Bombyx mori ,between Japanese(Fukuoka) Bombyx mandarina M.and Bombyx mori ,and between Bombyx mandarina M.and Japanese(Fukuoka) Bombyx mandarina M.were 0.52,0.63 and 0.58,respectively.Moreover,the genetic distance between Japanese (Fukuoka) Bombyx mandarina M.and Chinese (Shenyang) Bombyx mandarina M.was the smallest with a value of 0.48.Phylogenetic maps were also constructed in this research.
