摘要
目的 研究GPIb在凝血酶诱导的血小板活化过程中的作用。方法 FCM法检测SZ - 16的组织特异性 ,比浊法检测该单抗对血小板聚集的抑制作用 ,放射免疫法检测SZ - 16对凝血酶诱导的血小板活化的影响。结果 SZ - 16只与血小板结合 ,不与红细胞、白细胞和内皮细胞结合 ;血小板聚集实验表明 ,SZ - 16对凝血酶诱导的血小板聚集有抑制作用 ;放射免疫结果表明 ,SZ - 16抑制凝血酶引起的GMP140释放和TXB2的产生。结论 GPIb是血小板上的一种重要的凝血酶受体。
Objective To Study the function of GPIb in platelet activation induced by thrombin.Methods FCM examines the specificity of SZ-16. Turbidimetry examines the antibody inhibitory effect on platelet aggregation by exogenous agonist. Effect of SZ-16 to platelet activation induced by thrombin is examined by radioimmunoasay.Results SZ-16 could only bind to platelet, not to RBC?WBC and EC.Aggregation test showed SZ-16 could inhibite thrombin-induced platelet aggregation, and had not influence on ADP?Ristocetin and AA-induced platelet aggregation. Radioimmunoassay test showed SZ-16 could inhibit GMP140 release and TXB2 production induced by thrombin.Conclusion GPIb is an important thrombin receptor on platelet.
