摘要
目的 :了解化疗联合重组Endostatin对小鼠种植瘤的治疗作用 ,为肿瘤的免疫治疗提供条件。方法 :构建小鼠En dostatin原核表达质粒 ,在大肠杆菌表达小鼠Endostatin融合蛋白。分离纯化后 ,与丝裂霉素C联合应用 ,治疗种植在小鼠腿部肌肉内的小鼠肝癌H2 2肿瘤。观察瘤重 ,免疫组化检测肿瘤血管密度和肿瘤坏死情况。结论 :单纯应用重组Endostatin治疗种植瘤即产生明显的抑制作用 ,和化疗合用后 ,抑制作用更强 ,肿瘤血管密度减少 ,肿瘤细胞大量坏死。
Objective: To investigate the therapeutic effect of chemotherapy combined with recombinant endostatin on mice transplanted tumors. Methods: Mouse endostatin prokaryotic expression plasmid was constructed and it was expressed in E.coli. After purification, the recombinant protein was used with mitomycin C to treat mouse H22 heptic carcinoma transplanted in the leg muscle of mice. The weight of tumors were measured and vessel density was observed by immunochemistry and necrosis of tumor cells.Conclusions: The transplanted tumors were suppressed by recombinant endostatin alone and were highly suppressed after combined with chemotherapy.The density of vacularity decreased and the necrosis of tumor cells increased.
