摘要
目的 :为预测环孢素A(CSA)治疗再生障碍性贫血 (AA)疗效而建立体外药敏试验。方法 :采用微量甲基纤维素半固体培养粒—单核细胞集落 (GM CFU) ,并用CSA干预。结果 :体外 /体内 (S :敏感 ,R :耐药 ) :S/S11例 ,R/R10例 ,R/S :2 0例 ,与临床总符合率 5 1.2 % ,阳性符合率 10 0 % ,阴性符合率 3 3 .3 % ,试验敏感性 3 5 .3 5 % ,特异性10 0 %。结论 :CSA体外敏感试验与临床疗效有一定相关性 ,如体外敏感 ,则体内 10 0 %敏感 ,但存在一定假阴性。
Objective:To find a method of testing cyclosporin A(CSA) sensitivity for aplastic anemia (AA) in vitro. Methods: Methylcellulose semi solid cultures were used to assess GM-CFU colonies to estimate the sensitivity of AA to CSA. Results: There were 11 S (sensitive)/S (in vitro/in vivo), 10 R(resistant)/R, 20R/S in 41 Patients. In general, Positive and negative coincident rates of in vitro with thoses of in vivo, specificity, and sensitivity were 51.2%,100%,35.35%,100%,33.3%, resectively. Conlusion: The method gives a good predicitive value for the therapy of AA. If it is sensitive in vitro, rate of sensitivity in vivo is 100%, but there are some false negative.
