摘要
目的 建立眼角膜、结膜细胞体外培养的方法。方法 采用组织块培养法培养兔眼角膜基质及结膜成纤维细胞。结果 结膜成纤维细胞在培养 10天以后可以形成单层细胞。眼角膜成纤维细胞在培养瓶中培养 2周后可形成单层 ;2 4孔板培养 3d后细胞贴壁生长旺盛 ,10d形成单层。结论 眼角膜、结膜成纤维细胞组织块培养法是可行的 ,但在 2 4孔培养板上眼角膜成纤维细胞比在培养瓶中贴壁快而且容易生长。
Object To investigate the culture technique for corneal and conjunctival fibroblast.Methods The corneal and conjunctival fibroblast for rabbits were cultured by tissue culture.Result The conjunctival fibroblast of rabbit formed a monolayer and had a fibriform arragement after 10d culture.The corneal fibroblast of rabbit formed a monolayer and had a fibriform arragement after two weeks culture in culture flasks. The cells grew very well in 24-well culture plate for 3 days culture and had a fibriform arragement after 10d culture.Conclusion Two kinds of eye cells from rabbit can be cultured successfully.It is easier to culture corneal fibroblast in 24-well plate than in flask.
