摘要 目的 :研究多药耐药基因mdr1在口腔癌中的表达。方法 :2 6例未接受任何药物化疗的口腔癌患者标本 ,Tca8113,Tca8113 V10 0和Tca8113 V2 0 0细胞系 (分别耐药 10 0nmol和 2 0 0nmol) ,提取总RNA ,以pchp_1作探针进行斑点杂交 ,根据标准条计算出每个样本的mdr1拷贝数。结果 :2 6例口腔癌mdr1拷贝数范围为每微克 0 3× 10 5~5× 10 5总mRNA。其中 9例为不耐药 ,14例为中等耐药 ,3例高度耐药 ,耐药表达为 6 5 % ,与肿瘤大小及病理分级无关。结论 :口腔癌表达mdr1较高 ,临床要注意选择化疗方案。 Objective:The expression of mdr1 is associated with chemotherapeutic efficacy of cancer, some patients with oral cancer are resistant to chemotherapy, the authors therefore investigated the mRNA expression level of mdr1 in oral squamous carcinoma.Methods:Totally 26 samples were investigated in this study, and the total RNA of oral squamous cell carcinoma each sample were extracted and hybridized with probe pchp_1 .The level of mdr1 were compared with the cell line Tca8113, Tca8113/100 and Tca8113/200, which were sensitive, resistant to VCR 100 nmol and 200 nmol respectively.Results:The results showed that in the 26 patients, the mdr1 level ranged from 0.3×10\+5 to 5×10\+5. Among the 26 patients, 9 were drug_sensitive; 14 were low drug_resistant; and the other 3 were highly drug_resistant. The rate of drug resistance was 65%. The expression of mdr1 was not related with the pathologic differentiation and TNM classification.Conclusion:The multi_drug resistance of the malignant tumor to chemotherapy is the important factor of unsatisfied effect of chemotherapy, and this paper systemically studied on the mdr1 expression of the oral squamous cell carcinoma, and the relation with pathologic differentiation and TNM classification. The result is important for choosing the chemotherapeutic regime for patients.
