摘要
目的 建立适用于研究抗青光眼药物降眼压作用及保护视神经作用的兔青光眼模型。方法 将 2 1只兔随机分为Ⅰ、Ⅱ、Ⅲ及Ⅳ组 ,分别对其前房内注射复方卡波姆、甲基纤维素及复方甲基纤维素 ,结膜下注射地塞米松诱发青光眼 ,并对 4种药物诱发的青光眼模型进行观察。结果 Ⅰ组高眼压持续 2 0~ 5 0d ,平均眼压为 2 9~ 35mmHg(1mmHg=0 133kPa) ,眼压峰值为 37~ 4 5mmHg,青光眼模型成功率为 91 7%。Ⅱ及Ⅲ组模型有 10 %~ 2 0 %的兔眼眼压升高持续 3~ 4d ,如按眼压为2 2mmHg持续 1周的标准判断 ,Ⅱ及Ⅲ组模型均不理想。Ⅳ组眼压平均升高 3mmHg ,持续 7d ,亦为失败模型。结论 复方卡波姆诱发的兔青光眼模型具有引起眼压中度、稳定升高的时间长 ,方法简单 ,易于操作和控制等优点。可用于对青光眼性视神经视网膜损害的研究及对抗青光眼药物的研究 。
Objective To establish models of experimental glaucoma in rabbits to investigate the treatment of glaucoma. Methods Twenty one normal albino rabbits weighing 2 5 3 0 kg were studied The rabbits were divided randomly into 4 groups: Ⅰ, Ⅱ, Ⅲ and Ⅳ Anterior chamber injection of 0 1 ml of 0 3% carbomer (Ⅰ), 0 2 ml of methylcellulose (Ⅱ), 0 2 ml of compound methylcellulose (Ⅲ) and subconjunctival injection of 5 mg of dexamethasone (Ⅳ) were performed respectively on them to induce glaucoma The intraocular pressure (IOP) was monitored with a Tono pen XL tonometer several times prior to injection and 2 times per week after injection The basic IOP in every group served as control Results Experimental glaucoma occurred in eleven of twelve eyes in group Ⅰ and IOP lasted from 20 to 50 days The mean IOP levels were 29 35 mmHg (1 mm Hg = 0 133 kPa) and the peak IOP levels were 37 45 mm Hg The IOP in 2 out of 10 eyes in group Ⅱ increased to 22 50 mm Hg and lasted for 3 days The IOP of 1 out of 10 eyes in group Ⅲ increased to 25 40 mm Hg and lasted for 4 days The IOP in eyes of group Ⅳ increased only 3 mmHg in the mean and lasted for one week According to the standard, IOP above 22 mmHg for one week as the successful experimental glaucoma, group Ⅱ and Ⅲ were not the ideal ones and group Ⅳ was a failure Conclusion Experimental glaucoma model induced by carbomer has the advantages of producing moderate and sustained IOP elevation The model is easy to be carried out. It is simple, reliable, and useful for investigating optic nerve and retinal damage in glaucoma and testing the toxicity and efficacy of various therapies
