Correlation of the expression of p16, p53 and c-myc and cell proliferation with prognosis in gastric stromal tumor

目的 探讨胃间质瘤 (GST)中p1 6、p53、c myc基因蛋白及增殖细胞核抗原 (PCNA)与预后的关系。方法 应用免疫组化S P方法对 71例GST组织进行p1 6、p53、c myc基因蛋白和PCNA检测 ,并将PCNA的不同表达进行细胞增殖指数 (PI)分级。结果 71例GST中p1 6、p53、c myc基因蛋白阳性表达率分别为 :恶性 34 .8%、95 .7%、1 0 0 % ;交界性 45 .5 %、77.3 %、90 .9% ;良性 65 .4%、73 .1 %、65 .4%。p1 6蛋白表达PI分级Ⅰ、Ⅱ级明显高于Ⅲ、Ⅳ级(P <0 .0 1 ) ,存活期 <3年的明显低于 >3年的 (P <0 .0 1 )。p1 6蛋白随肿瘤分化程度和生存率降低 ,阳性表达率也逐渐降低 ;而p53、c myc蛋白及PI分级则相反。p1 6与p53、c myc蛋白及PCNA的PI分级有明显负相关性(P <0 .0 5)。结论 p1 6、p53、c

Objective To study the relation between the expression of p16, p53, c myc and proliferating cell nuclear antigen (PCNA) and prognosis of gastric stromal tumor (GST). Methods Expression of p16, p53 and c myc proteins was detected and PCNA labeling index was evaluated for cell proliferation using S P immunohistochemical method in 71 cases of GST. Results The rates of positive staining for p16, p53 and c myc were 34.8%, 95.7% and 100% for malignant GST,and 45.5%, 77.3% and 90.9% for borderline GST,and 65.4%, 73.1% and 65.4% for benign GST respectively. A significant correlation was found between low expression of p16, over expression of p53 and c myc, degrees of the tumor cell differentiation and survival rate (P<0.01). Conclusion Detection of p16, p53, c myc and PCNA is useful in assessment of GST differentiation and prognosis.
