卡介苗对鼠源性巨噬细胞株释放一氧化氮和IFN-γ的影响

摘要 目的 :观察卡介苗 ( BCG)对鼠源性巨噬细胞株 RAW2 64释放的诱导型一氧化氮 ( NO)合成酶 ( i NOS)及γ干扰素 ( IFN-γ)表达的影响。方法 :在鼠源性巨噬细胞株 RAW2 64培养液中加入BCG,Griess检测法测定 NO释放量 ,半定量 RT- PCR法检测 i NOS及 IFN-γ的表达。结果 :BCG添加后使巨噬细胞 NO产生量增加 ,IFN-γ、i NOS的 m RNA表达增强。结论 :在巨噬细胞吞噬、杀灭结核分枝杆菌时 IFN- Objective: To estimate bactericidal effect of NO on M.tuberculosis. Methods:BCG was added into RAW 264 culture, the quantity of released was determined NO through the Griess method and the expression of iNOS and IFN γ was detected through semi quantitative RT PCR technique.Results:The quantity of NO released from RAW 264 was increased and the IFN γ and iNOS mRNA expression was enhanced after adding BCG. Conclusion: IFN γ may activate the beactericidal effect of macrophage on M.tubercalosis.
