急性胰腺炎大鼠体内一氧化氮含量变化的意义

摘要 目的 :探讨急性胰腺炎 ( AP)大鼠体内一氧化氮 ( NO)变化规律及意义。方法 :将 64只Wistar大鼠随机分成对照组及阻断肝门后 1、3、6、1 2、2 4 h、N-硝基 - L-精氨酸 ( L- NNA)、L-精氨酸( L- Arg)实验组。用 5 %牛黄胆酸钠逆行注入法制成 AP模型 ,测定血和组织中 NO含量 ,并观察补充外源性 L- Arg的作用。结果 :1 h组血清和组织中 NO含量有一过性升高现象 ,显著高于其他各组 ( P<0 .0 5 ) ,然后逐渐下降 ,至 2 4 h已显著低于对照组 ( P<0 .0 5 )。结论 :AP时 NO有显著变化 ,NO在 AP早期有一过性升高现象 ,而后降至正常水平以下。适量的 NO对 AP具有一定的保护作用 ,补充外源性 L- Arg对 Objective:To study on the law of varied contents of endogenous endothelial vasodilator factor,nitrogen monoxide (NO), in the rat with experimertal acute pancreatitis (AP). Methods: 64 Wistar rats were devided into control group ,groups of hepatoportal block at 1, 3, 6, 12, 24 h, respectively, L NNA and L Arg experimental groups.The animal model with acute pancreatitis were made through retrospective injection of sodium taurocholate acid into pancreatocholecystic duct. Then the change of NO content in the blood and tissue was access with, and at the same time the effectiveness of exogenous supplement of L Arg was observed in the rats. Results: The seral and tissue contents of NO in the 1 h group were temporarily higher than other groups,having great significance ( P <0.05). But it reduced gradually, and were significantly lower than the control group in the 24 th hour ( P <0.05).Conclusion: The NO content in the rats with experimental acute pancreatitis varied a lot. It is temporarily elevated in the early stage, and then lowed down to below the nomal level. It implied that keeping NO content at suitable level is protective in the acute pancreatitis, and exogenous supplemnt of L Arg has great therapeutic effect.
