摘要 目的探讨以可吸收性珊瑚羟基磷灰石(Interpore500R)为细胞载体对骨髓基质细胞向成骨细胞分化、表型维持的影响。方法将生长状态良好的人骨髓基质细胞接种于Interpore500R上后行体外培养,通过细胞计数检测附着后的生长增殖特性、镜下观察附着情况及细胞形态,共聚焦显微镜下观察细胞分泌I型胶原情况,测定细胞内碱性磷酸酶含量变化。结果骨髓基质细胞接种后可继续增殖,并表达成骨细胞特性。结论可吸收性珊瑚羟基磷灰石具有促进骨髓基质细胞表达成骨细胞表型、合成细胞外基质的功能,是良好的细胞载体。 Objective To explore the effect of absorbable coral hydroxyapatite (Interpore 500R) used as cell culturing scaffold on the differentiation of human marrow stromal cells. Methods After inoculating the cells onto the surface of Interpore 500R, we surveyed the characteristic of proliferation by cell counting. Using scanning electronic microscope we observed celluar morphology. Staining collagen of I type (immunocytochemistry method) and observing the results under confocal microscope. Measured the content of cellular ALP. Results Human marrow stromal cells went on proliferating and started to differentiate to osteoblast after they attched on the material surface. Conclusion The absorbable coral hydroxyapatite is a good bone tissue engineering scaffold.
