摘要
以切花百合品种间杂交获得的胚龄为30~50d的幼胚作为外植体 ,进行离体培养基的筛选。结果表明 :基本培养基对萌发率、成苗率、生长势的影响从好到差顺序为MS>N6>SH>LS>Monnier>DCR>改良White;碳源种类对萌发的影响从好到差顺序蔗糖、甘露醇、白糖、葡萄糖、果糖 ,胚的成活情况与渗透压密切相关 ;生长素(NAA)不同浓度对幼胚成活的影响从好到差顺序为0.001>0.01>0.1>1>0,细胞分裂素(6-BA)不同浓度对幼胚成活的影响从好到差顺序为0.001>0.01>0.1>1>0。采用正交试验设计对幼胚萌发率及生长情况进行分析得出最优组合培养基为:糖60g·L-1+6 -BA0.1mg·L-1+NAA0.001mg·L-1+CH和VB6,基本培养基为MS附加琼脂10g·L-1,pH值为5.
The immature embryoes were used as the explants which were 30~50d after interspecific hybridization to screen in vitro culture media. The result indicated that the influence of the basic media on germination rate, seedling rate, growth potential was MS>N6>SH>LS>Monnier>DCR>Improved White. The effect of carbon sources on germination ranked from good to bad as sucrose, mannitol, white sugar, dextrose, fructose. The survival of embryoes had a closely relation with osmotic pressure. The effect of NAA concentration on immature embryo survival was 0.001>0.01>0.1>1>0; the effect of BA was 0.001>0.01>0.1>1>0. Based on the analysis of the immature embryo germination rate and growth in an orthogonal experiment, the best medium was sugar 60g·L-1, BA0.1mg·L-1, NAA0.001mg·L-1, basic medium MS with agar 10g·L-1 added CH and VB6, pH5.0.
