5个棉花纤维突变体胚珠和纤维的离体诱导 被引量:12

摘要 离体诱导不同类型的纤维突变体胚珠纤维发育 ,研究纤维突变体离体纤维发育的情况。结果表明 :不同基因型突变体的纤维离体发育各不相同 ,纤维诱导率和纤维生长量均较对照差。纤维突变体诱导产生棉纤维的能力依次是 TM- 1 >Li>N1>T- 5 86>H10 。无絮突变体在 Bt培养基上能产生绒状细胞团 ,很少有单细胞纤维的生长。离体培养纤维以同时含 GA3和 IAA的 Bt培养基生长较好 ,一般以 5μmol· L- 1IAA + 0 .5μmol· L- 1GA3的 Bt培养基较合适。不同突变体对 GA3和IAA的浓度组合有特殊性。 GA3对胚珠膨大的作用大于 IAA,并易产生愈伤组织。以 fl、T- 5 86愈伤组织的发生频率较高 ,并以 Bt+ 0 .5μmol· L- 1IAA+ 5μmol· L- 1GA3培养基产生愈伤组织频率最高。 The fiber induction and development from cotton ovules of fiber mutants in vitro were studied and analyzed. The results showed that there were great differences in the fiber development for genotypes, the fiber inducing percentage and the fiber quantity were less than TM 1. Ability of fiber inducement in vitro from ovule of different mutants can be put in order: TM 1>Li>N 1>T 586>H 10 .Although Fiberless Xu Zhou142 (fl) had not ability to differentiate fiber and fuzz in vivo, it could be induced to produce a lot of fiber like cells from epidermal cells in Bt medium. Some hormones, such as IAA and GA 3, were necessary to induce fibers in vitro, The Bt medium with 5.0μmol·L 1 IAA and 0.5μmol·L 1 GA 3 was better for fiber induction and the development. The effects of hormonal concentration might be differential in different fiber mutants. GA 3 could not only promote expansion of ovule, but also easily induce ovule into callus. Fiberless Xu Zhou142 (fl) and T 586 were easier for callus induction than the other mutants, they had hightest callus frequency in Bt medium with 0.5μmol IAA and 5.0μmol·L 1 GA 3.
