摘要
香榧种子胚乳DNA抽提是RAPD和AFLP分子标记构建遗传图谱的基础性工作。经多次实验 ,我们对CTAB法抽提香榧种子胚乳DNA进行了改进 ,即先将胚乳冷冻、抽提时加入 β 巯基乙醇、抽提过程中用高盐TE重悬DNA CTAB沉淀、最后用异丙醇沉淀DNA。OD2 60 /OD2 80 比值检测结果为平均 1 82 4 ,而且DNA得率也较高 。
It is a basic job in constructing the genetic graph of Torreya grandis to extract DNA from endosperm in its seeds.After several experimentations,some alteration was found in ertract DNA from endosperm in seeds by CTAB method:frozen endosperm in seeds in the beginning,added 2-ME in extracting DNA,suspended DNA-CTAB subsidence again by TE(higher salt)in extracting process,and subsided DNA by isopropanol in the end.The average was 1.824 in result of checking pureness of DNA.So the extracted DNA by this method can fulfil task of RAPD and AFLP analysis.
