芦荟治疗大鼠实验性变应性鼻炎的分子生物学研究 被引量:14

Molecular biological study of aloe vera in the treatment of experimental allergic rhinitis in rat

摘要 目的 :探讨芦荟 (aloevera)治疗大鼠实验性变应性鼻炎的作用机制。方法 :用卵清蛋白致敏大鼠制成变应性鼻炎动物模型 ,将芦荟经鼻给药 ,观察其行为学差异 ;用组织病理学方法观察鼻粘膜改变 ;用逆转录 多聚酶链反应 (RT PCR)对鼻粘膜和脾组织IL 2 ,IL 4mRNA进行定量检测。结果 :行为学得分 ,阳性对照组[(8.4 2± 1.0 6 )分 ]明显高于实验组 [(2 .0 2± 0 .4 2 )分 ]和正常对照组 (0分 ) ,其差异有极显著性意义 (P <0 .0 1) ;实验组鼻粘膜炎性反应明显轻于阳性对照组 ;实验组IL 2mRNA表达水平明显高于阳性对照组 ,其差异有极显著性意义 (P <0 .0 1) ;实验组IL 4mRNA表达水平明显低于阳性对照组 ,其差异也有极显著性意义 (P <0 .0 1)。结论 :芦荟通过改变TH1和TH2细胞因子基因表达 ,调节CD4+ Objective:To study the therapeutic mechanism of aloe vera in allergic rhinitis (AR). Method:Ovalbumin sensitized white rat used as animal models of AR were treated intranasally with aloe vera.At the end of treatment,the differences in the behavior science were observed;the changes in the nasal mucosa were studied by pathological;IL 2,IL 4mRNA in the nasal mucosa and spleen were used to do reverse transcriptive polymerase chain reaction (RT PCR). Result:The behavior science score of positive controls ( 8.42 ± 1.06 ) was higher than the experimental group ( 2.02 ± 0.42 ) and normal controls (0); inflammatory reactions in the experimental group nasal mucosa were remarkably relieved; the mean expression level of IL 2 mRNA in the experimental group was higher significantly than positive controls (P< 0.01 );but that of IL 4 mRNA was lower evidently (P<0.01).Conclusion: The aloe vera are suggested to be involved in the differentiation of CD 4 + lymphocytes, by means of regulating the expression of Th 1 and Th 2 cytokines.The results suggests that local aloe vera treatment was a selective and non traumatic method to treat the allergic rhinitis.
