MAPK参与调节猪卵母细胞和受精卵细胞周期的转变 被引量:3

摘要 从猪卵巢中获取卵母细胞,在体外成熟、体外受精和电激活后的不同时间采集样品,经裂解变性后利用蛋白电泳和蛋白免疫印迹技术,检测其中MAPK磷酸化的变化,并且用免疫荧光化学法观察ERK2的迁移.结果显示,猪卵母细胞中MAPK的量基本不变.体外培养前GV期猪卵内MAPK无磷酸化,培养 20 h后 MAPK开始发生磷酸化,30 h时进一步增加,36h时有所下降,40h时达到最高,一直到60h仍未降低.在卵母细胞成熟过程中,ERK2由胞外向胞内迁移,并分布于核区.猪卵母细胞电激活后18,20h MAPK磷酸化降低,几乎被灭活,但22h时开始上升.体外受精12h后MAPK完全去磷酸化,16h时重新磷酸化.以上结果表明,MAPK磷酸化/去磷酸化在猪卵母细胞MI向MⅡ转化、受精后原核的形成及第一次有丝分裂的启动等方面可能发挥重要的调节作用.
