人脑胶质瘤CDK4、CyclinD1和P16基因蛋白表达的免疫组织化学研究 被引量:7

摘要 采用检测人脑胶质瘤中 CDK4、Cyclin D1和 P16基因蛋白的表达情况 ,探讨 G1→ S期细胞周期调节蛋白在胶质瘤发生 ,发展过程中的作用。对 39例近期手术的胶质瘤标本和 8例瘤旁正常脑组织标本进行免疫组织化学检测。结果显示 :CDK4蛋白在良性、交界性胶质瘤和瘤旁正常脑组织中的表达差异没有显著性意义 (P>0 .0 5 ) ,在恶性胶质瘤中的表达却有明显地增高 ,与前两者比较差异均有显著性意义 (P<0 .0 5 )。 P16蛋白在恶性胶质瘤中的表达显著低于在瘤旁正常脑组织和良性、交界性胶质瘤中的表达 (P<0 .0 1) ,但在后两者中的表达差异没有显著性意义 (P>0 .0 5 )。 Cyclin D1蛋白在三者中的表达水平均较低且差异无显著性意义 (P>0 .0 5 )。结果表明 :CDK 4蛋白的表达增高和 P16蛋白的表达下调发生在胶质瘤的较晚期阶段 ,与胶质瘤的恶性变和恶性胶质瘤的发生有关 ,而 Cyclin D1蛋白的表达可能与胶质瘤的发生和发展无关。 In order to study the effect of G 1 S cell cycle regulating protein on the development and progression of glioma, CDK 4、Cyclin D 1 and P 16 gene protein expressions were examined in cerebral glioma. 39 cases of glioma samples and 8 cases of normal brain tissue samples adjacent to the tumer were detected by SABC immunohistochemical staining. The expression of CDK 4 protein and P 16 preotein showed no significant difference between benign or half benign glioma and normal brain tissue( P >0 05). In comparison with the above two tissues, the expression of CDK 4 protein in malignant glioma was higher and that of P16 protein was lower. The differences had obvious significance statistically( P <0 05 and P <0 01). The expression of Cyclin D1 protein was weak in the three tissues and had no significant difference( P >0 05). The higher expression of CDK 4 protein and lower expression of P 16 protein occur in the late stage of glioma and are involved in glioma development and malignant progression. But the expression of Cyclin D1 protein probably shows no relationship with the glioma dovelopment and progression.\;
