大白鼠颌下腺促性腺激素释放激素受体(GnRHR)mRNA的RT-PCR和原位杂交的研究 被引量:1

摘要 本实验采用 RT- PCR法探讨大白鼠颌下腺是否存在 Gn RH受体 m RNA,并用原位杂交法对其细胞定位进行了研究。结果显示 RT- PCR可扩增出大白鼠颌下腺 Gn RH受体 m RNA的特异性片段 ,其碱基数与设计的一致。原位杂交发现颌下腺浆液性腺泡上皮细胞、颗粒曲管、排泄管及分泌管上皮细胞内有 Gn RH受体 m RNA的杂交信号。信号物质分布于胞质内 ,胞核阴性。上述结果表明大白鼠颌下腺能合成 Gn RH受体 ,颌下腺产生的 Gn RH可作用于颌下腺的靶细胞 ,参与颌下腺生理功能的调节。 Using RT PCR and in situ hybridization we have studied whether gonadotropin releasing hormone receptor (GnRHR) mRNA is existed in rat submaxillary and its cellular localization The gene fragment from rat submaxillary GnRH receptor mRNA was amplified GnRHR mRNA hybridization signal was strong in glandular epithelial cells of serous acinus and ali gland ducts The signals only distributed in cytoplasm of all positive cells The results showed that GnRHR mRNA existed in submaxillary and was widely distributed in glandular epithelial cells with distributive pattern similar to those of GnRHR It suggests that the GnRH is produced by submaxillary and might bear the resposibility for modulating biological function of submaxillary \;
