大鼠肺泡巨噬细胞NOS表达的细胞化学和免疫细胞化学研究 被引量:1

摘要 用 NADPH-黄递酶细胞化学和 i NOS免疫细胞化学方法 ,结合图像定量分析 ,动态研究了 1μg/ml脂多糖诱导培养的大鼠肺泡巨噬细胞 i NOS的表达情况。结果发现 :脂多糖刺激 1小时染色反应强度与对照组相比无显著差异 ,6小时后明显高于对照组 ,随刺激时间延长 ,染色反应强度呈增高趋势 ,18小时后则趋于相对平稳 ;两种染色法显示的阳性细胞均以核周区染色反应为著。实验从形态学角度表明 :(1) NADPH-黄递酶细胞化学染色能够反映培养的大鼠肺泡巨噬细胞 NOS表达状况 ;(2 )脂多糖可诱导肺泡巨噬细胞 i NOS表达 ,其主要存在于核周区胞浆中。 Cytochemical and immunocytochemical methods in combination with image processing system were used for quantitative analysis. We investigated iNOS expression in rat alveolar macrophage in vitro after being given 1 μg/ml LPS. The results showed the intensity of stained reaction had no significant difference between LPS group and control group after one hour, however, the intensity of stained reaction was markedly increased in LPS group than that in control group after six hours. As time went on, the intensity of stained reaction became more obvious in LPS group until eighteen hours. The stained positive cells were located mainly in perinuclear zone both in cytochemical and immunocytochemical methods. The morphological results indicated that: (1) NADPH diaphorase cytochemical method could reflect NOS expression in cultured alveolar macrophage of rat in vitro; (2) LPS could induce iNOS expression in rat alveolar macrophage; the majority of iNOS expression was located in endochylemia of perinuclear zone.\;
